Antibodies’ CDR regions are crucial for their interaction with antigens. Humanization involves grafting these regions into human frameworks to reduce immunogenicity. This method, pioneered in the late 1980s by Greg Winter’s team, is now a mature and widely accepted strategy.

We offer humanization design services for antibodies and nanobodies derived from sources such as mice, rabbits, alpacas, and camels. Humanization strategy is as follows:

(1) Extract variable region sequences and predict structures using AI models.
(2) Human templates blast and CDR grafting: Select the optimal template combination based on homology, pharmacological properties, literature data, PI, etc.
(3) Score the key residues affecting antibody conformation, function, and stability. Apply back mutations, and generate humanized variant sequences. Predict PTM risks, and immunogenicity risks.
(4) Express and identify candidate humanized variants.
